Pramlintide was engineered with three proline substitutions specifically to suppress the fibrillation that makes native human amylin unusable, and that shows in the assay: reconstituted pramlintide is stable enough for a 24-h autosampler queue, which native amylin is not. The Cys2–Cys7 disulfide still reduces, giving the +2 Da open chain 0.9 min early. Thin coverage; the index has fewer than 50 pramlintide reports.
